Cathedral in the name of St. Nicholas the Wonderworker in Novogrudok opened in 1846 after the closing of the Franciscan Church, previously located in the building of the temple.

The Franciscan monks were invited to the Novogrudok Grand Duke Gediminas in 1323. In 1780 was built the Franciscan monastery, and with it, the majestic Church of St. Anthony, built with funds donated by Helena radziwiłł in the late Baroque style in the shape of a ship.

In 1831, "at the behest" of the monastery and the Church were closed and the Franciscans were forced to leave the territory of the Russian Empire.

In 1846 vacant majestic Church of St. Anthony was consecrated in the Orthodox Church of St. Nicholas the Wonderworker. In 1852 in Novogrudok was a large fire, which severely damaged the St. Nicholas Church. During the restoration works the building was substantially rebuilt and acquired Byzantine features inherent in Orthodox churches. The pediment on the facade was replaced with a bell tower. Orthodox iconostasis was painted in Moscow icon-painting workshop.

The Church has two altars dedicated to St. Nicholas the Wonderworker and Holy Martyr tsarina Alexandra.

In Novogrudok Nicholas Cathedral Orthodox shrines are stored: the icon of St. Nicholas the Saint of Myra and particles of the Holy relics
The icon of the Holy Trinity; the icon of the Mother of God "Joy of All who sorrow" (with Gresini). In the crypt of the Church built a Church of saints Kirill and Mefodii, teachers Slovenski.

After the restoration of Orthodox shrines in 1992, the Cathedral has the status of a Cathedral.
